My site has multiple affiliate links, and it works very well. I use Clickbank, Shareasale, and a few other networks. I also try to only promote products that I believe in.

It's also important to give your visitors some free information and reports, and not to try to make money off of every single spot on your site. I HATE when I see a work from home site that is covered in banners!

I find it easier to have a big site with lots of niches than to build lots of smaller sites. But whatever works for you, use it!
